Stem Cell-Derived Human Neurons in CNS Drug Discovery 07 Jun 2010: In this application note from Molecular Devices reproducible batches of human neurons are generated that will significantly improve and impact CNS drug discovery. The stem cell-derived neurons express all known markers typically associated with mature neurons and are functionally active. These neurons have been used in a recent screen to identify antagonists of a novel GPCR target implicated in schizophrenia. Molecular Devices

Cytotoxicity in Cells: Easy Determination Using Lonza ViaLight Plus and ToxiLight BioAssays on the SpectraMax L Microplate Luminometer 07 Jun 2010: The Lonza ViaLight® Plus and ToxiLight® Bioluminescent Cytotoxicity Assays exploit the cell characteristics of ATP levels and adenylate kinase release respectively, as cytotoxicity measures and enable detection using bioluminescent methods. This application note demonstrates that the SpectraMax L Microplate Luminometer and SoftMax Pro Software from Molecular Devices combine well with both assays providing a fast and sensitive combination of technologies in the field of cell death. Simplifying Peptide Bioanalysis 08 Apr 2010: This application note demonstrates that the Oasis PST μElution Method Development plate and protocol can greatly simplify extraction method development and rapidly identify an appropriate SPE sorbent and selective starting protocol for peptide bioanalysis. Such standardized approaches for determining optimal SPE recovery and concentration, and MRM-based LC/MS analysis may help to shorten time to market for peptide drugs. Waters

RapidFire® In Vitro ADME Applications 08 Apr 2010: This application note demonstrates the correlation between traditional LC-MS and RapidFire for a spectrum of ADME assays. The RapidFire® MS System is shown to perform in vitro ADME assays rapidly and without modifications to existing assay methods. Sample cycle times of approximately 6 seconds can also be achieved obtaining a 7 point IC50 curve in 42 seconds. BIOCIUS Life Sciences

Automation of Multiplexed Cell-Based Assay to Measure Induction of Gene Expression and Activity of Cytochrome P450 Isoform 3A4 08 Apr 2010: This application note demonstrates the automation of a cell-based multiplexed assay to explore how a drug can induce gene expression and inhibit activity of the same isoform, creating a useful tool to measure the stimulatory and inhibitory effects of a drug. Activity and induction of gene expression of CYP3A4 were measured using a luminogenic, cell-membrane permeable 3A4-specific substrate and a luciferin based reporter gene assay. Nephelometric Monitoring Growth of Candida albicans Using BMG LABTECH’s NEPHELOstar 08 Apr 2010: Laser nephelometry has been shown to be a reliable technique for the measurement of drug solubility in 96-well plate format. In this application note the effects of complexation on drug antimycotic activity is described. Laser nephelometry can distinguish between the concentration at which the drug just goes into or just comes out of solution. This is presented in phase solubility studies. BMG LABTECH

Optimizing Donor Number for Consistent Pooled Human Liver Microsomes 08 Apr 2010: Human liver microsomes (HLMs) are commonly used as an in vitro reagent for the study of human CYP in the metabolism of a drug candidate. This application note presents an analysis of the CYP activity distributions for over 300 characterized (HLM) samples and predicts the variability in pooled CYP activity as a function of donor number. An optimal donor number based on observed variability can be selected. BD Biosciences

Live Cell Imaging of Drug Effects on Golgi Morpholgy Using the CELLview™ Cell Culture Dish 01 Mar 2010: This application note by Greiner Bio-One shows that performing live-cell imaging experiments in parallel in CELLview™ dishes with four compartments allows a direct comparison of the speed and timing of drug effects on the Golgi apparatus. GalNAc-GFP cells can be easily imaged at multiple positions without cell death or focus problems. Greiner Bio-One GmbH
